Protease specificity and protease activity can be assayed by measuring the cleavage rate of protein substrates. One method is to incubate the protease with a protein substrate that has been modified with a large number of fluorophores. Since many fluorophores on the same molecule result in fluorescent quenching, the cleavage of these molecules causes an increase in fluorescence.
