Protein Mods offers a variety of molecules for incorporation into your protein, including a selection of fluorophores as well as agarose bead, thiol and enzyme conjugations.
Fluorophores: useful for protein visualization and quantitation. Fluorophores absorb light at a characteristic range of wavelengths and emit light at a longer range of wavelengths. Common applications involve fluorescent microscopy, flow cytometry, and a variety of assays for microtiter plates.
- Fluorescein: Excitement by blue light causes fluorescein to emit green light.
- Tetramethylrhodamine: Excitement by green light causes tetramethylrhodamine to emit yellow and red light (550 nm max excitation, 580 max emission).
- Cy5.5: Excitement by green or red light causes Cy5 to emit far red light (680 nm max excitation, 700 max emission). CY5.5 is compatible with LICOR imaging systems using the 700 channel.
Sulfhydryl (thiol): useful for protein conjugations. Thiolated proteins can bind to gold surfaces by a covalent gold-thiol bond (biosensors and SPR applications). A thiolated protein can be conjugated to another protein by a heterobifunctional crosslinker such as SMCC (reacts with amines on one end and with thiols on the other).
Horseradish Peroxidase (HRP): useful for protein quantification and signal amplification. With proteins conjugated to HRP, measurement of peroxidase activity indicates protein concentration. For western blotting, use of an HRP-conjugated primary antibody means there is no need for a secondary antibody. This not only saves time, but also may reduce the background associated with non-specific binding of the secondary antibody. We will perform this service for any protein. However, we only remove unconjugated peroxidase from IgG antibodies.
Agarose bead (Sepharose): useful for co-precipitation studies, affinity chromatography and immunodepletion. In these applications, proteins conjugated to agarose beads are typically exposed to complex mixtures. Molecules that interact with the protein on the bead can then be isolated from the rest of the mixture through centrifugation or chromatography.
